Genetics and the nephron.

چکیده
There have been phenomenal advances in our understanding of renal biology over the last 20 years through our ability to define the genetic mutations causing kidney disease in children. This review will take you through a trip down the nephron and highlight how these conditions may present to the paediatrician and the molecular basis for their biological effects.

A Microdosimetry model of kidney for nephrotoxicity due to internal radiation therapy
Introduction: We describe a nephron-based dosimetry model for renal toxicity for radiopharmaceutical therapy suited to the nonuniform activity distribution of radiopharmaceutical. In radiopharmaceutical therapy, renal toxicity is observed while whole-kidney and renal cortex absorbed dose values are below toxicity thresholds established by external beam and targeted radiope ptid...
متن کاملMITF – A controls branching morphogenesis and nephron endowment
Congenital nephron number varies widely in the human population and individuals with low nephron number are at risk of developing hypertension and chronic kidney disease. The development of the kidney occurs via an orchestrated morphogenetic process where metanephric mesenchyme and ureteric bud reciprocally interact to induce nephron formation. The genetic networks that modulate the extent of t...
متن کاملTowards the integration of genetic knowledge into clinical practice.
The past three decades have seen a revolution in molecular biology and genetics that have changed the way we define disease, diagnose it, understand pathogenesis, initiate new treatments and assess individual responsiveness to intervention. The 'new genetics' has made its biggest impact on monogenic or 'rare' diseases, although its impact is increasingly being felt in the polymorphic so-called ...
متن کاملThe proximal tubule phenotype and its disruption in acute renal failure and polycystic kidney disease.
In light of recent developments in the fields of genetics, molecular, cell and developmental biology, the kidney is receiving increasing attention as a model system for organ development and human diseases. Gene disruption experiments have provided evidence for the essential role of a number of proteins in the earliest phase of nephron development, but very little is known about the identity of...
متن کاملEvolutionary Nephrology
Progressive kidney disease follows nephron loss, hyperfiltration, and incomplete repair, a process described as "maladaptive." In the past 20 years, a new discipline has emerged that expands research horizons: evolutionary medicine. In contrast to physiologic (homeostatic) adaptation, evolutionary adaptation is the result of reproductive success that reflects natural selection. Evolutionary exp...
